I have the following line in $HOME/.muttrc:

set signature="~/bin/sigrot.sh|"

The pipe at the end of the command tells mutt to read the signature line
from the output of the script $HOME/bin/sigrot.sh. Here are the contents 
of sigrot.sh:

SIGFILE=$HOME/.signature
/usr/games/fortune -s | tee $SIGFILE
